چکیده
Abstract Introduction Perineuronal nets (PNNs) surrounding fast-spiking, parvalbumin (PV) interneurons provide excitatory:inhibitory balance within cortical circuits. This is impaired in several disorders that are also associated with altered diurnal rhythms, yet few studies examined rhythms of PNNs or PV cells. Methods We measured the intensity and number cells labeled Wisteria floribunda agglutinin (WFA) oxidative stress marker 8-oxo-deoxyguanosine (8-oxo-dG) rat prelimbic medial prefrontal cortex (mPFC) at Zeitgeber times (ZT) ZT0, 6, 12, 18. To examine changes inhibitory excitatory inputs to cells, we GAD 65/67 vGLUT1 puncta apposed without PNNs. Whole-cell slice recordings fast-spiking was conducted determine ratio alpha-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid receptor:N-methyl-D-aspartate receptor (AMPA:NMDA) ZT18 vs. ZT6. Finally, PV/PNN containing orthodenticle homeobox 2 (OTX2), which maintains PNNs, assessed. Results Relative intensities PNN labeling were increased dark compared light phase. The 8-oxo-dG decreased from ZT0 all times. There more on ZT6, but no puncta. an AMPA:NMDA OTX2 showed a strong trend toward increase ZT6 ZT18, differences non-PV-containing Conclusion Diurnal fluctuations alter balance. Detailed understanding how these regulated should new insights into treatments for diseases impacted by disturbances sleep circadian rhythms. Ongoing examining downstream signaling after removal. Objectives: The purpose of this study is to investigate the alterations of the hypothalamic-pituitary-adrenal axis hormones, especially salivary cortisol and dehydroepiandrosterone (DHEA) in perinatal depression. Methods: 44 patients with depression and 217 normal subjects in perinatal period were included in this study. Edinburgh Postnatal Depression Scale(EPDS) and Beck Depression Inventory I...
متن کاملDevelopmental disruption of perineuronal nets in the medial prefrontal cortex after maternal immune activation
Maternal infection during pregnancy increases the risk of offspring developing schizophrenia later in life. Similarly, animal models of maternal immune activation (MIA) induce behavioural and anatomical disturbances consistent with a schizophrenia-like phenotype in offspring. Notably, cognitive impairments in tasks dependent on the prefrontal cortex (PFC) are observed in humans with schizophren...
متن کاملStructural Changes in the Medial Prefrontal Cortex and Anterior Cingulate Cortex of Dehydroepiandrosterone-Induced Wistar Rat Model of Polycystic Ovarian Syndrome
Introduction: Polycystic ovary syndrome (PCOS) is a complex endocrine disorder in women that is associated with an increased risk of infertility. This study aims to evaluate the neurobehavioral and neurochemical changes along with the associated changes in the medial prefrontal cortex (mPFC) and anterior cingulate cortex (ACC) of the dehydroepiandrosterone (DHEA)-induced PCOS model rats. Metho...
متن کاملParvalbumin-positive basket interneurons in monkey and rat prefrontal cortex.
Differences in the developmental origin and relative proportions of biochemically distinct classes of cortical neurons have been found between rodents and primates. In addition, species differences in the properties of certain cell types, such as neurogliaform cells, have also been reported. Consequently, in this study we compared the anatomical and physiological properties of parvalbumin (PV)-...
